Have you inserted a disk into drive D:\? If you have either your drive is no good or you don't have the proper drivers installed.
When you insert the pen drive does the OS (Operating System) do anything? You should get a prompt asking you what you want to do.
Has the pen drive been formatted? If you right click and choose properties do you have any options?
What OS (Windows XP, Windows Vista, Windows 7)?
